Isothermal titration calorimetry (ITC) is a technique used to quantitatively study the interactions of various biomolecules. It directly measures the heat released or absorbed during the binding of biomolecules. ITC is the only technique able to determine all binding parameters simultaneously in one assay. ITC can determine the affinity of the binding partner in the natural state without modification of the binding partner by fluorescence labeling or immobilization techniques.

ITC has several advantages over other techniques for studying biomolecular interactions: it does not require any chemical modification or immobilization of any binding partner; the molecular weight of the reactant is not restricted, and it is not affected by spectroscopic properties of reactants.
