1. The role of PEG in protein precipitation/protein purification
Non-ionic polymers such as PEG were first used to purify immunoglobulins and precipitate some bacteria and viruses. In recent years, they have been widely used in the separation and purification of nucleic acids and enzymes. The advantages are: (1) mild operating conditions, not easy to cause denaturation of biological macromolecules; (2) high precipitation efficiency, using a very small amount of PEG to precipitate a considerable number of biological macromolecules; (3) organic polymers are easy to remove after precipitation.
2. The role of PEG in protein concentration
PEG acts as a water-absorbing agent because of its strong water-absorbing properties. The water-absorbing agent can also be made into a solution with a concentration of 30%-40%, and the dialysis bag containing the protein solution can be put into it. After the water absorbing agent is used, it can be dried in a thermostat or dried naturally, and it can still be used again. It is mainly used to replace the protein buffer, and a dialysis bag is sufficient, and no special equipment is required.
3. PEG can modify proteins and liposomes
PEG modifiers are pH neutral, non-toxic, water-soluble polymers, with high hydrophilicity, good biocompatibility and blood compatibility, and no immunogenicity. Therefore, the structure modification of polyethylene glycol can improve the properties of the drug.
In addition, PEG can also modify the carrier liposomes used for drug delivery. Traditional liposomes and immune liposomes are easily recognized and taken up by cells of the reticuloendothelial system (RES), resulting in a very short blood circulation half-life (usually less than 30 min), which is eliminated before reaching the target organ, so their application is very limited. If the hydrophilic polymer molecule polyethylene glycol is introduced on the surface of the liposome membrane, a layer of hydration film can be formed on the surface of the liposome, which can cover the hydrophobic binding site on the surface of the liposome, hinder the access of plasma components to the liposome, thereby reducing the recognition and uptake of liposomes by RES, and prolonging the blood circulation time of liposomes.
4. PEG can promote cell fusion
PEG is a synthetic agent, which has high fusion and can effectively promote the fusion of plant cells and animal cells. In addition, the fusion activity of each batch is consistent, the fusion efficiency is stable, and the cytotoxicity is low.
5. PEG can be used as a surfactant.
The solubility between PEG and many organic compounds is also very good. It is because of this unique property that one of its main functions is to act as a solvent or surfactant to help the components with different chemical properties to mix together uniformly. At the same time, polyethylene glycol has a strong interaction with water, so the polyethylene glycol added to these products helps keep it moist. In addition, some daily chemicals require a certain viscosity, and polyethylene glycol comes in handy at this time. Since low molecular weight polyethylene glycols are viscous liquids, they can significantly increase the viscosity of these daily chemicals.
